Sounds like a very interesting project! I know a good bit about acoustics, from a practical point of view, as I operate as a hobby an old analog recording studio. The project you suggest is indeed possible, though based on various acoustic properties at the site of the "sample" you may get very different results. Theoretically, the difference from a calibration value may get you within a tolerable error.
Currently I have been doing a great deal of work with audio capture, playback, and synthesis. I can send you a sample app for iOS or Android that captures audio and finds the pitch using a Fast Fourier Transform. I guess it is sort of like a "guitar tuner," but I do a bit more with the data than that.
