HIV genotyping is important for subtype detection. Also it is used for genotyping drug resistance monitoring. A drug efficacy is measured using this techniques. I have been using this technique for regular monitoring of HIV positive individuals.

Cells were infected with two primary HIV-1 isolates, AG204 and mSTD104, and a cell line-adapted strain, Ba-L. Six days after infection, cells positive for HIV-1 antigens were detected by IFA using a fluorescence microscope. Percentage of cells judged to be positive for IFA are shown. I worked in the Virology department, Gunma University, Japan under supervision of Dr. Hiroo Hoshino (A senior author of this article).

Lymphocyte proliferation assay (LPA) measures the ability of lymphocytes placed in short-term tissue culture to undergo a clonal proliferation when stimulated in vitro by a foreign molecule, antigen or mitogen. CD4+ lymphocytes proliferate in response to antigenic peptides in association with class II major histocompatibility complex (MHC) molecules on antigen-presenting cells (APCs). This proliferative response of lymphocytes to antigen in vitro occurs only if the patient has been immunized to that antigen, either by having recovered from an infection with the microorganism containing that antigen, or by having been vaccinated. Therefore, some normal individuals may not respond to a given antigen, but most people will respond to at least one of several common microbial antigens.

The nucleic acids of various viruses encode surface proteins (e.g.hemaaglutinin (HA) of influenza virus) that agglutinate red blood cells (RBC) of a variety of species. The reaction of viral hemagglutinins with red blood cells results in a lattice of agglutinated cells which settle irregularly in a tube or microtiter well. Unagglutinated cells settle in a compact button. This process is known as hemagglutination and it is done in Influenza detection.

I had received a Scholarship from the Japanese Student Service Organization for Advance Medical Research, Gunma University Graduate School of Medicine, Japan. I learned there HIV culture in GFP SL-III lab. Using this technique we can diagnose critical HIV positive clients.HIV drug resistance and propagation of other viruses like SERS-CoV-2 which is the causal agent of COVID-19. Also other respiratory (Influenza), enteric (Rota), bloodborne(HIV, HCV, Dengue) pathogens.
